    A probable aculeacin A acylase from the Ralstonia solanacearum GMI1000 is N-acyl-homoserine lactone acylase with quorum-quenching activity

    <p>Abstract</p> <p>Background</p> <p>The infection and virulence functions of diverse plant and animal pathogens that possess quorum sensing systems are regulated by <it>N</it>-acylhomoserine lactones (AHLs) acting as signal molecules. AHL-acylase is a quorum quenching enzyme and degrades AHLs by removing the fatty acid side chain from the homoserine lactone ring of AHLs. This blocks AHL accumulation and pathogenic phenotypes in quorum sensing bacteria.</p> <p>Results</p> <p>An <it>aac </it>gene of undemonstrated function from <it>Ralstonia solanacearum </it>GMI1000 was cloned, expressed in <it>Escherichia coli</it>; it inactivated four AHLs that were tested. The sequence of the 795 amino acid polypeptide was considerably similar to the AHL-acylase from <it>Ralstonia </it>sp. XJ12B with 83% identity match and shared 39% identity with an aculeacin A acylase precursor from the gram-positive actinomycete <it>Actinoplanes utahensis</it>. Aculeacin A is a neutral lipopeptide antibiotic and an antifungal drug. An electrospray ionisation mass spectrometry (ESI-MS) analysis verified that Aac hydrolysed the amide bond of AHL, releasing homoserine lactone and the corresponding fatty acids. However, ESI-MS analysis demonstrated that the Aac could not catalyze the hydrolysis of the palmitoyl moiety of the aculeacin A. Moreover, the results of MIC test of aculeacin A suggest that Aac could not deacylate aculeacin A. The specificity of Aac for AHLs showed a greater preference for long acyl chains than for short acyl chains. Heterologous expression of the <it>aac </it>gene in <it>Chromobacterium violaceum </it>CV026 effectively inhibited violacein and chitinase activity, both of which were regulated by the quorum-sensing mechanism. These results indicated that Aac could control AHL-dependent pathogenicity.</p> <p>Conclusion</p> <p>This is the first study to find an AHL-acylase in a phytopathogen. Our data provide direct evidence that the functioning of the <it>aac </it>gene (NP520668) of <it>R. solanacearum </it>GMI1000 is via AHL-acylase and not via aculeacin A acylase. Since Aac is a therapeutic potential quorum-quenching agent, its further biotechnological applications in agriculture, clinical and bio-industrial fields should be evaluated in the near future.</p

    Description of local dilatancy and local rotation of granular assemblies by microstretch modeling

    AbstractThis study investigates the microstretch continuum modeling of granular assemblies while accounting for both the dilatant and rotational degrees of freedom of a macroelement. By introducing the solid volume fraction and the gyration radius of a granular system, the balance equations of the microstretch continuum are transformed into a new formulation of evolution equations comprising six variables: the solid volume fraction, the gyration radius, the velocity field, the averaged angular velocity, the rate of gyration radius, and the internal energy. The bulk microinertia density, the averaged angular velocity, and the microgyration tensor at a macroscopic point are obtained in terms of discrete physical quantities. The bulk part and the rotational part of the microgyration tensor are proposed as the two indices to measure the local dilatancy and local rotation of granular assemblies. It is demonstrated in the numerical simulation that the two indices can be used to identify the shear band evolution in a granular system under a biaxial compression

    COMPARISON OF KINESIO TAPING AND SPORTS TAPING IN FUNCTIONAL ACTIVITIES FOR COLLEGIATE BASKETBALL PLAYERS: A PILOT STUDY

    The purpose of this study was to identify the effects of kinesio taping and sports taping for collegiate basketball players in functional activities. Seventeen collegiate basketball players were recruited in particapitate this study and divided into three groups(Kinesio taping group, sports taping group and control group) in random. The ankle range of motion, plantarflexor muscle strength and endurance, vertical jump, and dynamic balance were measured before and after taping applied. The results was showed significantly increasing in ankle plantar-flexion range for Kinesio taping group(p=.03). There were no remarkable difference in the other measurements. In conclusion, the Kinesio Taping would not restriction the ankle plantar-flexion range. In future, we may recruited more subjects to identify the effect of Kinesio taping in functional activities for collegiate bsketball players

    A fault-tolerant triple-redundant voice coil motor for direct drive valves: Design, optimization, and experiment

    AbstractA direct drive actuator (DDA) with direct drive valves (DDVs) as the control device is an ideal solution for a flight actuation system. This paper presents a novel triple-redundant voice coil motor (TRVCM) used for redundant DDVs. The TRVCM features electrical/mechanical hybrid triple-redundancy by securing three stators along with three moving coils in the same frame. A permanent magnet (PM) Halbach array is employed in each redundant VCM to simplify the system structure. A back-to-back design between neighborly redundancies is adopted to decouple the magnetic flux linkage. The particle swarm optimization (PSO) method is implemented to optimize design parameters based on the analytical magnetic circuit model. The optimization objective function is defined as the acceleration capacity of the motor to achieve high dynamic performance. The optimal geometric parameters are verified with 3D magnetic field finite element analysis (FEA). A research prototype has been developed for experimental purpose. The experimental results of magnetic field density and force output show that the proposed TRVCM has great potential of applications in DDA systems

    Hospital admissions for respiratory system diseases in adults with intellectual disabilities in Southeast London:a register-based cohort study

    BACKGROUND: Intellectual disability (ID) carries a high impact on need for care, health status and premature mortality. Respiratory system diseases contribute a major part of mortality among people with ID, but remain underinvestigated as consequent morbidities. METHODS: Anonymised electronic mental health records from the South London and Maudsley Trust (SLaM) were linked to national acute medical care data. Using retrospective cohort and matched case–control study designs, adults with ID receiving SLaM care between 1 January 2008 and 31 March 2013 were identified and compared with local catchment residents for respiratory system disease admissions. Standardised admission ratios (SARs) were first calculated, followed by a comparison of duration of hospitalisation with respiratory system disease between people with ID and age-matched and gender-matched random counterparts modelled using linear regression. Finally, the risk of readmission for respiratory system disease was analysed using the Cox models. RESULTS: For the 3138 adults with ID identified in SLaM, the SAR for respiratory system disease admissions was 4.02 (95% CI 3.79 to 4.26). Compared with adults without ID, duration of hospitalisation was significantly longer by 2.34 days (95% CI 0.03 to 4.64) and respiratory system disease readmission was significantly elevated (HR=1.35; 95% CI 1.17 to 1.56) after confounding adjustment. CONCLUSIONS: Respiratory system disease admissions in adults with ID are more frequent, of longer duration and have a higher likelihood of recurring. Development and evaluation of potential interventions to the preventable causes of respiratory diseases should be prioritised

    Suppress vibration on robotic polishing with impedance matching

    Installing force-controlled end-effectors on the end of industrial robots has become the mainstream method for robot force control. Additionally, during the polishing process, contact force stability has an important impact on polishing quality. However, due to the difference between the robot structure and the force-controlled end-effector, in the polishing operation, direct force control will have impact during the transition from noncontact to contact between the tool and the workpiece. Although impedance control can solve this problem, industrial robots still produce vibrations with high inertia and low stiffness. Therefore, this research proposes an impedance matching control strategy based on traditional direct force control and impedance control methods to improve this problem. This method's primary purpose is to avoid force vibration in the contact phase and maintain force-tracking performance during the dynamic tracking phase. Simulation and experimental results show that this method can smoothly track the contact force and reduce vibration compared with traditional force control and impedance control
